Nicotinamide riboside, often abbreviated as NR, is a form of vitamin B3 and a precursor of nicotinamide adenine dinucleotide, or NAD. NAD is a coenzyme found in all living cells and is involved in energy metabolism, redox reactions, DNA repair and cellular signaling.
NR occurs naturally in small amounts in foods such as milk and yeast. In supplements, it is generally used in a concentrated and standardized form.
NAD exists mainly in two forms: NAD+ and NADH. These forms continuously convert into one another as part of cellular energy production. NAD+ accepts electrons, while NADH donates them in metabolic pathways that help generate ATP.
NAD is also required by enzymes such as sirtuins and PARPs. Sirtuins are involved in nutrient sensing, stress responses and gene regulation, while PARPs participate in DNA repair. Because NAD levels may decline with age, researchers are investigating whether NAD precursors such as NR can influence aspects of healthy aging.
After ingestion, NR is absorbed and converted through several enzymatic steps into nicotinamide mononucleotide, or NMN, and then into NAD. The precise pathway may vary between tissues.
NR therefore does not act as NAD itself. It serves as one of several building blocks the body can use to replenish the cellular NAD pool.
Animal studies have reported that NR can increase NAD levels and may influence mitochondrial function, metabolic regulation and cellular resilience. Human studies have also shown that NR can raise NAD-related metabolites in the blood.
However, evidence for specific clinical outcomes—such as improved physical performance, slower aging or prevention of disease—remains limited. Results differ according to dose, study population and duration.
NR is discussed in longevity research because NAD-dependent enzymes are involved in several hallmarks of aging. These include mitochondrial dysfunction, genomic instability, altered nutrient sensing and cellular stress responses.
Increasing NAD availability may support these pathways, but NR should not be considered a proven anti-aging treatment. It is one potential component of a broader strategy that also includes exercise, sleep, a balanced diet, metabolic health and avoidance of smoking.
NR has generally been well tolerated in short- and medium-term clinical studies. Possible adverse effects may include digestive discomfort, headache or fatigue. Long-term safety data are still limited.
People who are pregnant, breastfeeding, undergoing cancer treatment or taking medication should discuss supplementation with a physician. The same applies to anyone with chronic liver, kidney or metabolic disease.
